Isis mBAND (High Resolution Multicolor
Banding)
 MetaSystems proprietary high
resolution multicolor banding technique mBAND provides precise information on
intra-chromosomal rearrangements and exact break-point mapping. mBAND answers
the questions that mFISH leaves
unanswered. Providing highly reproducible, high resolution pseudocolor bands, mBAND
is the method of choice for the detection of intrachromosomal rearrangements, i.e.,
inversions, insertions and deletions. With its unique approach mBAND reveals a
banding pattern with approx. 500 bands in the normal haploid human karyotype. The Isis mFISH and mBAND software module
provides a variety of tools for the precise analysis of any mBAND specimen,
including user-trainable color classifiers for mBAND (for details see the
mFISH section).

mBAND is based on region-specific partial chromosome
paints (PCP) combined with quantitative color ratio analysis. Each PCP is labelled using a
unique fluorochrome combination and partly overlaps with its neighbouring one. The
resulting fluorescence intensity pattern along the chromosome axis shows a continuous
change of color ratios. Pseudo colors can be assigned to chromosome sections of similiar
color ratios giving rise to a reproducible color banding pattern that does not depend on
chromosome condensation.
To obtain i.e. a high resolution color banding of chromosome 5, seven partial
chromosome paints were used. 25 color bands are achieved which corresponds to a resolution
of approx. 500 bands for the total haploid chromosome complement. The ratio analysis
effectively multiplies the initial resolution of the probe kit by a factor of 3 to 4. The
striking advantage of mBAND over "color bar code" approaches that use
individually labelled YACs or BACs is obvious: in order to obtain the same number of color
bands, with the unique mBAND technique the complexity of the probe cocktail as
well as the number of fluorochrome combinations is at least three times less as compared
to the color bar code approach.
