XCyte mBAND Multicolor Chromosome Banding Kits*

mBAND

The unique mBAND technique from MetaSystems couples combinatorial labelling of specifically designed probes and a sophisticated fluorescence intensity ratio analysis. The highly reproducible false color banding pattern gives a resolution of up to 500 bands per haploid human genome. The color banding patterns can then be used to investigate intra-chromosomal rearrangements such as inversions, insertions and deletions.

The kits are prepared from overlapping microdissected and fluoro-chrome labelled sections of chromosomes. As with the 24XCyte mFISH probe kit, the XCyte mBAND kits use 5 different fluorochromes, where 4 of them are incorporated directly, whilst biotin is detected using a Streptavidin Cy™5-conjugate (see B-tect).

Compared to other banding techniques, mBAND gives much greater scientific information. The pattern is nearly independent from chromatin condensation, and user trainable classifiers make the system adaptable to different preparation conditions.

mBAND preparations can be imaged with the efficient FISH imaging system Isis. The captured images can then be analyzed using our mFISH/mBAND module to give precise information on structure and banding localization.

Reliable and simple probe kit protocols coupled with easy-to-use imaging software makes mBAND the perfect assay for the analysis of intrachromosomal aberrations.

mband-schemes_small.jpg (7096 Byte)The XCyte mBAND probe kit contains a cocktail of different chromosome section painting probes. Each probe is labelled with 1 of 5 different fluorochromes (labelling schemes). Since the probe sections are partly overlapping each other, and unique color combinations can be identified with the mFISH / mBAND module of the FISH imaging software Isis, the final banding resolution is much higher than the number of partial chromosome paints used (approx. 500 bands per genome).
